We are looking for a highly-motivated Research Engineer to be in charge of the newly created DECODERS platform, supported by the Labex who am I? and hosted by the Institut Jacques Monod (CNRS/Université de Paris, France).
Several research groups at Université de Paris aim at understanding the molecular bases of genomic processes such as transcription, replication and repair, in a variety of model systems and physiological (or pathological) situations. As a support for their research, the DECODERS platform aims to implement a common methodological pipeline for spatially-resolved proteomics and transcriptomics at individual genomic loci. The chosen strategy relies on proximity biotinylation approaches coupled to CRISPR-based targeting to specific DNA sequences, an efficient and versatile method for the identification of the protein and RNA species enriched at given genomic or cellular locations (Nat. Methods 15:433 & 15:437; Mol. Cell 75:875; Cell 178:473).
The recruited engineer will:
- set-up the platform and implement the workflow in a defined set of pilot experimental models (e.g. repeated sequences in mammalian cultured cells and budding yeast);
- use this optimized workflow for characterizing locus-specific interaction networks in several alternative models, in collaboration with the different teams of the consortium. The engineer will notably contribute to design the experimental setups, generate the molecular and cellular tools, train the users to the workflow and realize certain steps of the procedure.
Proteomic analyses will be performed with the ProteoSeine facility (Institut Jacques Monod).
The applicant must have a PhD in biological or biochemical sciences and demonstrable experience in molecular biology and biochemistry. Previous expertise in proximity proteomics (BioID, TurboID, APEX) and/or genome editing (CRISPR-Cas9) will be greatly appreciated. Strong organizational and team working abilities, as well as verbal and written communication skills in English will be required.
The DECODERS platform is hosted by Institut Jacques Monod (IJM), one of the main centers for basic research in biology in the Paris area, within the Université de Paris campus in the Paris left bank.
