Job Description

Mammalian cells must recycle membranes and nutrients after degradation in lysosomes to maintain homeostasis and conserve resources. This recycling activity is particularly crucial in phagocytic cells, where engulfed material is degraded and reused rather than expelled. Phagocytosis, a receptor-mediated internalization and digestion of objects larger than 0.5 microns, leads to the formation of a phagosome that matures through a series of fusion and fission with the endosomal compartments to generate a phagolysosome. This implies important remodeling of the phagolysosome, with fragmentation and fission steps, and the resulting vesicles then replenish the pool of lysosomes, in a step called phagosome resolution. How phagolysosomes resolve however still lacks detailed characterization. While the fate of phagosomes and their internalized cargo depends on the surface receptors initially engaged, the mechanisms by which phagocytic receptors modulate phagosome terminal resolution remain poorly understood.
We will investigate these critical steps of phagosomal resolution using micron-sized lipid particles functionalized with phagocytic ligands and decorated with fluorescent tracers for pH and proteolytic activity. This design will enable innovative real-time analysis of biochemical and mechanical processes underlying phagosome maturation and resolution in living cells.
Your Work Environment
The project is supported by the 80 I PRIME (Projet de recherche inter-instituts multi-équipes) interdisciplinary CNRS programme (http://www.cnrs.fr/mi).
The PhD candidate will be co-supervised by Florence Niedergang (Team « Biology of Phagocytes, Infection and Immunity» at Institut Cochin, www.institutcochin.fr) and Jacques Fattaccioli (Institut Pierre Gilles de Gennes, CNRS, ENS Chemistry Department, Sorbonne Université), in close collaboration with Jean-Maurice MALLET and Blaise DUMAT (CNRS, ENS, Sorbonne Université).
The principal working place will be Institut Cochin, which is one of the biggest biomedical French Research Center located in the center of Paris. The other laboratories are located in Institut Pierre-Gilles de Gennes, a leading institute dedicated to microfluidics and its applications, and the Chemistry department of ENS/PSL, a top tier French university and research center.
The 3 laboratories are located in the Latin Quarter in Paris, within 15 min walking distance.
