Description du sujet de thèse
Design and develop new photobiocatalysis and enzyme engineering methodologies as part of the research project “Light-driven asymmetric enzymatic transformations using fatty acid photodecarboxylase” (ANR GreenCatFAP/BIAM).
Contexte de travail
The Aix Marseille Institute of Biosciences and Biotechnology (BIAM) is a Joint Research Unit (UMR) comprising around 100 permanent employees and 50 non-permanent employees (fixed-term contracts, doctoral and post-doctoral students, interns). The UMR is located on the CEA Cadarache site and on the Luminy Faculty of Science campus. The position is assigned to the Environment, Bioenergy, Microalgae, and Plants team.
Skills: - Master's degree in biochemistry or organic chemistry - Knowledge of protein biochemistry and protein physicochemistry - Knowledge of chemical synthesis - Knowledge of health and safety regulations - Design of experimental devices - Drafting of scientific documents, reports, and publications - Mastery of presentation techniques (oral and written) and meeting facilitation - Adaptation of educational content to the level and expectations of the audience - Good organizational and adaptability skills in a complex environment - Rigorous and autonomous work ethic - Ability to work in a team and participate in the life of the laboratory - Fluency in English
Activities - Develop, test, and optimize the technical options required for the project:
a) heterologous expression in E. coli, purification and characterization of FAP and its homologues;
b) development of high-throughput LC-MS screening for monitoring photo-induced radical additions;
c) directed mutagenesis/iterative evolution cycles to improve yield and stereoselectivity. - Write and update technical protocols, ensure their monitoring and quality; keep a rigorous laboratory notebook. - Exploit and present the results of analyses (written reports, oral presentations). - Participate in the drafting of funding applications and publications. - Supervise students (M1/M2 internships) and ensure technology transfer. - Apply and enforce health and safety rules in the field. - Monitor scientific and technological developments (photobiocatalysis, enzymology, radical chemistry).
