Description du sujet de thèse

Development of protein-based biotechnological tools through de novo design, directed evolution and protein engineering in the context of studying the molecular mechanisms of synaptic transmission and mechanobiology.

Activities:
Implementation of de novo protein design methods. Design of directed evolution strategies mainly using phage display. Implementation of strategies (creation of banks, screening, selection, etc.) for the selection of clones with the desired properties. Implementation of methods for the validation of isolated clones using biochemical, biophysical and imaging methods. Engineering of protein tools using chemical or genetic methods for use in a cellular environment. Validation of tools in cellular models using imaging, proteomics and biochemistry.
Supervision and training.

Expected skills:
General knowledge of chemistry and biology, basic concepts in mathematics, statistics and physics. Health and safety rules relating to the handling of toxic products, contaminants and genetically modified organisms (GMOs), The scientific field of structure, Mastery of experimental biology techniques, Knowledge of the principles and use of specific equipment in the field: incubators, spectrophotometers, thermocyclers, plate washers, microscopes, HPLC, SPR, ITC, etc. Using IT tools for data recording and equipment control. Communicating and managing relationships with internal and external contacts. Drafting technical procedures. Methodological and technological monitoring. Development and optimisation of methods and protocols.

Contexte de travail

Created on 1 January 2011, the Interdisciplinary Institute for Neuroscience (IINS) is a joint research unit affiliated with the CNRS and the University of Bordeaux. With 170 staff members divided into 12 teams, IINS combines multidisciplinary expertise to study the molecular and cellular mechanisms involved in brain activity. The Institute develops innovative methods in imaging, chemistry, physiology and computer science. As part of Bordeaux Neurocampus, IINS is one of the major players in the neuroscience community in Aquitaine.
