Job Description

Thesis Subject

HTLV-1 (Human T-lymphotropic virus type 1) is a virus that infects certain immune cells, known as CD4+ T lymphocytes. It causes a chronic inflammation of the nervous system called TSP/HAM and an aggressive cancer known as adult T-cell leukemia/lymphoma (ATL). Although it is different from HIV, HTLV-1 shares common mechanisms with HIV for replicating within cells, particularly the way its genetic material (viral RNA) leaves the cell nucleus with the help of a protein called CRM1. During this process, the viral RNA is monitored by the cell's defenses and must associate with other host proteins to form a protective and functional structure (vRNP) that is essential for infection. The background of this project is partly described in one of the team's most recent publications (Prochasson et al., 2025, NAR).
The primary objective of this thesis project is to understand the respective roles of the cellular protein CRM1 and the viral protein Rex in the formation of this vRNP.
The second objective is to characterize the functions of these vRNA-associated proteins throughout the viral cycle (from nuclear export to viral particle production).
The project involves conducting bench-top experiments (biochemistry, molecular biology, and cell culture) to achieve the objectives described above. Bioinformatics analyses will also be required following high-throughput approaches.

Your Work Environment

This project is part of the RicoReC program, funded by the ANRS and the Ligue contre le Cancer (see thesis project description).
The candidate will join the PRIO team, which focuses primarily on the study of post-transcriptional regulation in the contexts of viral infection and carcinogenesis.
The group is part of the Laboratory of Cell Biology and Modeling, which aims to characterize the molecular basis underlying the organization and functioning of cellular processes under normal and pathological conditions. It is based at the École normale supérieure de Lyon, a leading French research and teaching institute (LBMC, ENS de Lyon, UMR5239, U1293, Université de Lyon).
