Job Description
Thesis Subject
Atypical teratoid/rhabdoid tumors (AT/RT) are aggressive pediatric tumors that arise in the central nervous system in very young children. They are caused by mutations in the SmarcB1 tumor suppressor gene, which encodes a protein in the SWI/SNF chromatin remodeling complex. AT/RT tumors are characterized by significant changes in the epigenome and cellular metabolism. The goal of this thesis project is to characterize the dynamics of epigenome evolution over time in an AT/RT model based on SmarcB1 loss-of-function coupled with metabolic alterations in human brain organoids, using integrative computational approaches. The PhD student will be responsible for preparing samples for epigenomic analyses (ATAC-Seq, Cut&Run) and performing computational analyses. He or she will be integrated into two research teams: one based at the CRCT for the computational aspect (https://www.crct-inserm.fr/annuaire/vera-pancaldi/) and the other based at the CBI for the experimental aspect (https://cbi-toulouse.fr/equipe/davy/).
We are seeking a highly motivated, independent, and meticulous candidate with a master's degree in biology or bioinformatics. Expertise in computational epigenome analysis is required, and prior experience in cell culture would be a plus, as would familiarity with modeling concepts. Fluency in English is required to present results in frame of the PEPR Cell-ID or other scientific events.
Your Work Environment
The PhD student will be co-supervised by Vera Pancaldi (CRCT INSERM) for the computational component and by Alice Davy (CBI CNRS) for the experimental component. The Center for Integrative Biology (CBI) is one of the leading centers for basic research in biology in Toulouse; it is located on the Rangueil campus of the University of Toulouse. The Toulouse Cancer Research Center (CRCT) is a major cancer research hub located on the Oncopôle campus. Both institutes are easily accessible by public transportation.
