4D investigation of cellular interaction in a Drosophila model of ATRT brain tumours
Your Work Environment

On our 5-hectare campus in the heart of Paris (15th arrondissement), within a multicultural and stimulating environment, 3,000 people work together to fulfil the Pasteur Institute's ambitions
Within the 'Structures and Signals in the Neurogenic Niche' unit, you will work on a specific project focusing on the interactions between tumour cells and host tissue in a paediatric AT/RT-type tumour model in Drosophila.
Your responsibilities will be as follows:
Activity 1: Design and lead, as a specialist, the development of the various methodologies required to carry out the project
- Construction of dedicated genetic tools
- Implementation of complex genetic crosses
- Planning and carrying out immunolabelling experiments and other detection techniques (RNA FISH) on dissected brains
- Planning and carrying out transcriptional analysis experiments (Targeted DamID, single-cell RNA-seq)
- Acquiring confocal microscopy images: fixed samples and live tissues with real-time imaging
Activity 2: Process the data
- Image analysis (segmentation, quantification)
- Statistical analysis of quantitative data
Activity 3: Disseminating and promoting the results
- Generation of representative figures
- Poster and oral presentations at national and international conferences
Required skills:
Knowledge
- Handling and genetics of Drosophila
- Principles of immunolabelling
- Confocal microscopy
- Critical analysis, interpretation and presentation of results
- Working proficiency in English (reading, speaking, writing), enabling you to work in an English-speaking environment
Practical skills
- Dissection of adult Drosophila brains
- Standard molecular biology techniques (cloning, DNA and RNA extraction)
- Operation of a confocal microscope
- Proficiency in statistical software (principles and software such as Prism)
- Proficiency in the basics of image analysis and associated software (Fiji)
- Proficiency in IT tools (Word, Excel, databases, etc.)
