Missions
The candidate will carry out chemical syntheses on a microwave peptide synthesizer, purify the peptides obtained by chromatography, characterize them by NMR, UV-Vis and Fluorescence and study their coordination with Cu(ll) and Cu(l) ions using a range of spectroscopic techniques (EPR, NMR, CD, UV-Vis, Fluorescence) as well as their ability to model the catalytic activity of the LPMO protein, with a view to understanding its mechanism of action and developing synthetic mimetics.
Activités
- Automated synthesis of peptide polymers using a microwave-assisted automatic synthesizer:
Preparation of the synthesis protocol and programming of the apparatus,
Preparation of the solutions required for automated synthesis,
Checking that the automated synthesis stage is running smoothly.
- Peptide purification (flash chromatography, HPLC-MS in semi-preparative and preparative modes)
- Physico-chemical characterisation of peptides (NMR, UV-vis, Fluorescence)
- Inorganic synthesis of copper peptide complexes and physico-chemical characterisation (EPR, NMR, UV-vis, Fluorescence)
- Drafting of progress reports and presentation of results at group meetings (and possibly at scientific events)
- Technical control and maintenance of the peptide synthesizer
Compétences
- Organic and inorganic synthesis
- Mastery of classic spectroscopic characterisation techniques (UV-vis, NMR, etc.),
- Computer skills (word processing, spreadsheet) and software associated with characterisation techniques,
- Organisation and management of working time in order to carry out experiments within set deadlines,
- Ability to work as part of a team.
Contexte de travail
Most of the work will be carried out at the CNRS Coordination Chemistry Laboratory (UPR 8241) in the 'Alzheimer's and amyloid' team led by Dr. Christelle Hureau as part of an ANR grant. The project will benefit from the team's infrastructure, the host laboratory and the platforms available in Toulouse.
