The project that will be entrusted by the post-doctoral fellow is to develop a novel screening assay for drug discovery in the treatment of Freidreich's ataxia (FA). FA is a neurodegenerative and cardiac disease caused by defect in the expression of frataxin, a mitochondrial protein involved in the assembly of iron-sulfur clusters. Iron-sulfur (Fe-S) cluster are protein cofactors involved in a multitude of essential biological processes: ATP production, Krebs cycle, protein synthesis and maintenance of genome integrity. Their synthesis is achieved by a machinery comprising 7 proteins in a complex multistep process stimulated by frataxin. Based on recent achievements in the laboratory in the reconstitution of the Fe-S clusters assembly machinery, the project is to use a reconstituted machinery with purified proteins to screen chemical libraries for the discovery of molecules replacing frataxin.


The Post-doctoral fellow will be in charge of the production of the recombinant proteins constituting the machinery, its reconstitution with metal ions and the development of the screening assay under anaerobic conditions. The hits of the screens will be validated using several biochemical assays (Fe-S cluster assembly, sulfur transfer) and then tested in a drosophila FA models, in collaboration with Paris-Diderot University.


- Highly motivated candidate for drug discovery with a strong background in biochemistry, recombinant protein production and/or pharmaco-chemistry

- Rigorous, well-organized with acute skills in communication with the supervisor

- Specific skills in one or more of these areas: metallo-proteins, handling of anaerobic conditions using a glove box, redox biology and pharmacology will be appreciated.

- Writing of articles, communication of results and possibly trainee supervision

Work Context

I2BC is a CNRS, CEA and Paris-Saclay university mixt unit, recently created in the South-Paris area that gathers cell biologists, biochemists and biophysicists (about 700 researchers). The Post-doc will integrate the "redox biology and diseases" team that is a part of the Cell Biology department. It is an internationally recognized group studying redox biology, specially the mechanisms of Fe-S cluster assembly, and has made recent achievements in the discovery of the biochemical function of frataxin. The post-doc will work within a team of about 10 researchers, specifically on Friedreich's ataxia.
