Missions

This project aims to determine the deregulation of DNA damage checkpoints in high-grade serous ovarian cancer (HGSOC) and differential sensitivity to corresponding pharmacological inhibitors, in relationship with their genetic contents (driver mutations), with the expectation to provide a predictive value.

Activités

- Generate stable fallopian tube non-transformed cells and HGSOC cancerous cell lines genetically modified and expressing DNA damage checkpoint sensors and various proteins (fluorescent reporters,..) in a constitutive and/or in an inducible manner.
- Analyze the regulation and properties of DNA damage checkpoints during cell cycle progression in absence or under exogenous genotoxic stresses
- Transiently express these sensors into patient-derived ovarian tumor cells (ascite primary cultures).
- Perform molecular and cell biology experiments (DNA cloning, cell culture, transfection and/or lentiviral infection, genome editing by CRIPSR/Cas9: Knock-Out and Knock-In).
- Perform video-microscopy experiments on living cells using functional perturbation assays (drugs).
- Quantitatively analyze cellular events on time-lapse video-microscopy image series.
- Data presentation: laboratory meetings, internal and external seminars in English.

Compétences

- Most importantly, good interpersonal skills and ability to work as part of a team.
- At least 6 mounths full-time experience in a research laboratory.
- Thoroughness in carrying out experiments.
- Excellent organizational skills.
- Solid skills in molecular and cell biology: DNA cloning, culture and maintenance of mammalian cell lines, transfection/infection, genetic modification by CRISPR/Cas9.
- Fluorescence microscopy skills (on fixed and/or live cells).
- Experience in biological image analysis.
- Confident to speak in English.

Contexte de travail

Our team is part of the UMR9019 CNRS research unit "Genome Integrity and Cancer" located at the Institut Gustave Roussy (subway line 14) - Université Paris-Saclay, a leading cancer research center in Europe. The Institute has various on-site platforms for live-cell imaging (video, confocal, spinning disk, super-resolution microscope), intravital imaging in animals, flow cytometry, bioinformatics and genomics. The successful candidate will benefit from a stimulating scientific environment with several research groups working on DNA integrity and replication - repair mechanisms.

Contraintes et risques

Lentiviral infection will be carried out in the P3 containment laboratory.
Our institute provides mandatory training for access to and use of the P3.
