Missions
The project focuses on the characterization of the role of Fanconi anemia proteins, a rare genetic disease predisposing to leukemia, in the biogenesis of ribosomes and protein synthesis. We wish to identify a) the biochemical and molecular origins of the alterations in ribosome biogenesis that we have recently described in the cells of patients (Gueiderkh et al., Sci Adv, 2021); and, b) their consequences in terms of mRNA translation (translatome).
The experiments will be conducted on in vitro and ex-vivo cell models (mouse bone marrow cells) and will use techniques for analyzing the expression and activity of proteins and mRNAs: immunofluorescence, flow cytometry, immunoprecipitation, western blot, RNAseq from total cell extracts and polysome profiles.
Activities
The candidate will work in F. Rosselli's team and will have to (a) participate in the design and implementation of experiments (b) manage the organization of these experiments, (c) adapt experimental protocols , (d) autonomously carry out the quantitative analysis of the data including the appropriate statistical tests, (e) present the results obtained during laboratory meetings, (f) participate in the writing of manuscripts for the publication of the results, ( g) be able to supervise young researchers (M2/PhD)
Skills
The candidate must have experience in a research laboratory and great rigor in carrying out experiments. At the experimental level, advanced skills in cell biology (culture and maintenance of mammalian cell lines, transfection), molecular/biochemical biology, fluorescence imaging and cytometry are required. At the fundamental level, a good knowledge of transcription and translation processes is also required.
Work Context
Our team is part of the UMR9019 CNRS "Genome Integrity & Cancers" research unit located at the Gustave Roussy Institute. The unit and the institute have the necessary equipment for the smooth running of the experiments. The candidate will benefit from the expertise of a scientific environment of the Unit and will participate in the development of a new team project.
