Missions

As part of the ANR-DFG TreatProCDG project, the post-doctoral student in glycosciences will work on the analysis of glycans, the cellular environment, golgian trafficking and pH. The international project focuses on the role played by TMEM165 in the regulation of Ca/Mn ion homeostasis and golgi pH via potential interactions with the V-ATPase complex. The aim is to develop a treatment for CDG patients suffering from golgian pH deficiency using manganese supplementation.

Activités

I-Specific and principal activities - Cell biology techniques: Conservation and maintenance of fibroblast cell lines from CDG patients, stable and transient transfections, confocal microscopy and fluorescence analyses of the glycoproteome, imaging of living organisms. - Analysis of glycans and glycoproteins using classical glycomics approaches (mass spectrometry), metabolic labelling using non-natural sugar precursors and click chemistry (SiaNAl) and biochemistry (Western-blot and lectin-blot). - Development of a treatment for congenital glycosylation diseases linked to proton transport by manganese supplementation. II - Associated activities - Supervision of trainees (Bachelor's and Master's degrees) - Participation in international meetings - Funding applications

Compétences

- Knowledge General knowledge of biology and mastery of the basics of cell biology. Knowledge of glycobiology would be appreciated. II- Skills Cell biology skills required. Technical skills in biochemistry (Western-blot and lectin-blot) and a good knowledge of glycobiology would be an advantage. Fluency in scientific English (reading and writing). Ability to analyse and present information. Drafting articles for publication in international peer-reviewed journals and literature reviews. III-Abilities (Personal skills) Motivation, diligence, dynamism, innovation and rigour Scientific curiosity, reliability in the execution of work and creativity Ability to work both independently and in collaboration within a research

Contexte de travail

The Structural and Functional Glycobiology Unit is a multi-disciplinary research unit of around 150 people with an international reputation, where the research carried out is centred around a unifying theme: glycobiology. The studies carried out aim to understand the structure-function relationships of glycoconjugates. The person recruited will work in the 'Molecular mechanisms of glycosylation in health and diseases' team led by Dr François Foulquier. This team is interested in the structure-function relationships of glycoconjugates in healthy and pathological situations of genetic diseases of glycosylation (GCD). The young researcher's work will take place in building C9, on the campus of the University of Lille's Faculty of Science and Technology, avenue Mendeleïev in Villeneuve d'Ascq.
