Missions

Postdoctoral fellowship in brown algal molecular genetics

Brown algae (Phaeophyceae), or brown seaweeds, are multicellular macroalgae that belong to the larger eukaryotic Stramenopile supergroup. The ANR project BrownLincs aims to deepen the understanding of the molecular processes underlying brown algae development and acquisition of multicellularity through an analysis of the functional regulatory roles of long non‐coding RNAs (lncRNAs), key actors in cellular regulation across the Eukarya domain of life.

The candidate will delete selected lncRNA loci in the brown algal model organism Ectocarpus using CRISPR methodology. The phenotypes of the resulting mutants will then be analysed at both the morphological and molecular levels, the latter involving both transcriptomic analysis and chromatin analysis using ChIP-seq.

Activités

The project will involve maintenance and growth of wild type and mutant Ectocarpus strains, application of CRISPR-Cas9 methodology to generate new mutant lines and phenotypic characterisation of newly created mutant lines. Phenotyping will employ a range of different approaches including morphometry, cell biology approaches, transcriptomic and epigenetic approaches. Protocols are available in the host laboratory for all these approaches.

Compétences

The candidate should have experience with standard molecular biology and cell biology (microscopy) approaches and experience working with CRISPR methodology and/or handling brown algae in culture. Standard bioinformatic skills, for example the capacity to handle transcriptomic data, would also be appreciated.

Contexte de travail

The host institute, the Station Biologique de Roscoff (SBR, http://www.sb-roscoff.fr/about-roscoff-marine-station/missions.html), is a centre of marine biology and oceanographic research situated in the town of Roscoff on the north coast of Brittany (France) about 60 km east of Brest. The SBR has about 200 permanent employees.

The postdoctoral fellowship is financed by the "Agence Nationale de la Recherche" (ANR) project BrownLincs. The project will involve strong collaborative interactions with the l'Institut de Biologie de l'Ecole Normale Supérieure (IBENS) in Paris and the Institut Génétique & Développement (IGDR) in Rennes.
