Missions
Metallic nanoparticles activated or not by an external stimulus (laser, alternating magnetic field) can generate effects on the immune system that are favourable in certain tissue repair or anti-cancer therapy strategies. The aim will be to assess the immunomodulatory effects of metallic nanoparticles activated or not by external fields in three-dimensional models of atherosclerotic plaques or fibrous tumours. The focus will be on the immune infiltrate as a function of the physical properties of the extracellular matrix for different types of nanoparticles/stimuli.
Activités
- Setting up a 3D model of atherosclerosis and fibrous tumours and studying its mechanical properties.
- Incorporating immune cells into these 3D models and studying their behaviour.
- Screening the effect of nanoparticles on these 3D models, with or without exposure to external stimuli.
- Set up multivariate analyses and machine learning tools to screen the immune microenvironment in these models.
Compétences
As part of an ANR project, we are looking for a post-doctoral researcher. The PhD should preferably be obtained in 2024, with in-depth knowledge of biophysics or bioengineering or nanomedicine. Desired experience in :
- cell culture and 3D models (organoids)
- nanoparticles and their interaction with the biological environment and/or external excitation fields
- microfluidics
- immunology
Contexte de travail
The new multidisciplinary NABI unit (Nanomedicine, Extracellular Biology, Integratome and Innovations in Health) on the Saint Germain Campus of Université Paris Cité (45 rue des Saints Pères, 75006 Paris) has been created on 1 January 2025 and brings together around 70 physicists, biologists, pharmacists, engineers and doctors, including around 30 permanent staff, under the supervision of CNRS, INSERM and Université Paris Cité. The scientific objectives of NABI are to develop new therapies based on biological nano-objects (extracellular vesicles) or synthetic nano-objects (activatable metallic nanoparticles) for the delivery of personalised therapies for pathologies that are difficult to treat. The study of nano-objects in biological fluids requires trans-disciplinary approaches (physics, engineering, cellular and molecular biology, immunology, etc.) right through to clinical practice and dedicated technical and analytical resources (super-resolution microscopy, nanoprobes, microfluidics, etc.) as well as systematic multimodal and multi-scale exploration, the analysis of which calls on AI.
These innovative methodologies are developed within NABI and the industrial integrator IVETh, a technology hub dedicated to the characterisation, bioproduction and engineering of extracellular vesicles and nanovectors for diagnosis and personalised therapies. The post doctoral researcher will join the multidisciplinary 'Nanobiotherapies' team as part of the ANR NanoHypMPI project.
