Missions
As part of an INSERM project in collaboration with the teams of G. Salgado and Y. Hashem in Bordeaux, E. di Giorgio in Udine, the single molecule section of the LPENS Multi-Scale Physics of Life team (J. Valle-Orero, JF Allemand, and V. Croquette, in close collaboration with J-B Boulé of the MNHN) is working on the role of G-quadruplexes (G4) in the Kras promoter and their links with transcriptional regulation of the gene in cancer cells. The single molecule part of the consortium is involved in using magnetic tweezers to determine the kinetics of formation of the three potential G4s that may form on this promoter and the possible interaction between the G4s and with proteins interacting with the promoter and likely to have an effect on the G4s. To do this, we are using a protocol developed in the laboratory that allows G4s to be formed in a double-stranded hairpin context, as described in the reference: https://doi.org/10.1093/nar/gkab306
We are looking for an engineer to conduct this research.

-Measurement of G4 structure formation kinetics using magnetic tweezers as the main task.
-Molecular biology to set up experiments as a secondary task.

Compétences
Experience in single molecule manipulation is required (optical or magnetic tweezers, but preferably magnetic). The successful candidate must also have the necessary skills in molecular biology to construct the hairpins required for the experiments. The successful candidate may also be required to purify proteins that are likely to interact with the structures formed in J-B Boulé's group at the MNHN.
Contexte de travail
Under the supervision of the group's permanent staff, the successful candidate will conduct experiments to determine the formation times of different G4s and some of their mutants, and to observe the effect of the presence of oxoguanines at certain positions. The effect of proteins interacting with the promoter will be studied to determine their influence on the stability of G4s.
