Missions
At the Institut de Chimie-Physique (ICP) in Orsay, you will join the “Dynamics and Function of Redox Proteins and Oxidative Stress” team, which is studying the role and mechanisms of a multimeric redox protein complex essential to the functioning of the innate immune system. Using an approach combining experimental biochemistry and artificial intelligence tools, you will help to elucidate the structure-function relationships of this membrane complex, with the aim of developing innovative strategies to modulate its activity. Your main mission will be to design, produce and characterize protein chimeras, exploiting structural modeling tools to test their ability to influence the functioning of this enzymatic system.
Activités
- Use of artificial intelligence tools to design chimeric proteins in silico (AlphaFold, sequence alignments, etc.).
- Implementation of molecular biology and biochemistry techniques for the expression and purification of recombinant proteins.
- Biochemical and biophysical characterization of chimeras (purity, structure, stability, activity)
- Functional enzymatic tests to assess the impact of modifications
Compétences
- Mastery of structural modeling tools such as AlphaFold and sequence alignment techniques to guide protein engineering
- Experience in recombinant protein expression (prokaryotic or eukaryotic systems)
- Expertise in protein purification and characterization: FPLC, SEC, SDS-PAGE, BN-PAGE, western blot, UV/Vis spectroscopy, enzymatic assays, stopped-flow.
- Skills in formulation or use of lipid nanoparticles would be an asset
- Proficiency in data processing tools (Excel, scientific analysis software, etc.)
- Good command of scientific English (written and spoken)
- Rigor, autonomy, organizational skills
- Ability to work in a team and supervise trainees
Contexte de travail
You will work at the Institut de Chimie-Physique (ICP), located on the Université Paris-Saclay campus, in a high-level scientific environment.
You will join the “Biochemistry of redox and oxidative stress proteins” team, headed by Laura Baciou, which has all the equipment required for the production, purification, characterization and functional study of proteins.
