Job Description
Missions
Understanding the molecular mechanisms of translation of leaderless messenger RNAs in the archaeon Saccharolobus solfataricus.
The successful candidate will primarily use molecular biology and biochemistry—including structural biology, particularly cryo-EM—to characterize translation initiation complexes assembled on leaderless messenger RNAs.
Activity
Use of in vitro translation assays to characterize the influence of the 5'UTR region and the aS26 protein on translation efficiency in the context of a messenger RNA with a (potentially modified) 5' untranslated region (5'UTR) and a leaderless sequence.
Initiation complexes formed on leaderless messenger RNAs will be isolated from cell extracts and then characterized by cryo-electron microscopy to determine the three-dimensional structures of these complexes at high resolution.
Your Profil
Skills
The candidate (M/F) must hold a Ph.D. in biology, chemistry, or biophysics, have a strong background in cryo-EM and the purification of macromolecular complexes, and have at least one year of experience following completion of their dissertation.

The project will be conducted within the “Translation Mechanisms” team led by Emmanuelle Schmitt and Yves Mechulam at the “Structural Cell Biology” (BIOC) laboratory at École Polytechnique, https://portail.polytechnique.edu/bioc/en/recherche/translation-mechanisms. The BIOC laboratory is a joint research unit of the CNRS and École Polytechnique, located on the Palaiseau campus and fully equipped to carry out the project. In addition, the laboratory has privileged access to CIMEX (Interdisciplinary Center for Electron Microscopy at the École Polytechnique), which is equipped with a 300 kV Titan Themis electron microscope fitted with a Falcon 3 direct electron detector.
