Missions

To assist in the development and characterization in the pluripotent and differentiated state ( notably in organoids)of human induced pluripotent stem cell (hiPSC) lines carrying mutations causing neurodevelopmental or renal pathologies.

Activités

- maintenance of pluripotent iPSC cell lines
- cell differentiation, in 2D and 3D (organoids)
- characterization by immunofluorescence on cells and on cryosections of organoids, quantitative imaging, FACS, western blot and qRT-PCR analyses.
- depending on the evolution of the project: CRISPR/Cas9 approaches for the generation of new mutant iPSC lines

Compétences

- Strong experience in cell biology, including cell culture and immunofluorescence/immunohistochemistry is required.
- Previous experience with induced pluripotent stem cells (iPSCs) culture and differentiation into organoids would be appreciated
- Experience in microscopy and/or image analysis would be desirable but
training will be available on site (ImagoSeine facility)
- Ability to communicate and work in an international team is essential (good command of the English language).
- Organizational skills are required.
- Computer skills will be appreciated.

Contexte de travail

The engineer will join the team "Non conventional functions of nuclear pores" (https://www.ijm.fr/research-topics/research-groups/doye/?lang=en) which is part of the Institut Jacques Monod (https://www.ijm.fr). He/she will be supervised by Valérie Doye, DR1 CNRS and will also benefit from the expertise of the engineers of the enSCORE organoid platform (https://www.ijm.fr/plateformes-et-services/plateformes/enscore-vf/).
The Institut Jacques Monod includes about thirty teams conducting research on genome and chromosome dynamics, cell dynamics and signaling, development and evolution. These teams use quantitative biology and modeling approaches, and their research areas, although focused on fundamental research, frequently have an impact on the understanding of mechanisms underlying various pathologies. The Institute is located in the Buffon building on the Paris Rive Gauche campus of the Université Paris Cité. It offers a rich and multidisciplinary environment, as well as access to different experimental approaches within the laboratories and technological platforms, to ensure an adequate development of the research.
