Missions

The engineer will work on the StuDySARCOMERE ERC project and study the nano-architecture of adult and developing sarcomeres in Drosophila using a DNA-PAINT super-resolution microscope combined with oligonucleotide-labeled nanobodies. This will provide a better understanding of how sarcomeres assemble and mature. The candidate will work within an international team and collaborate with biophysicists and developmental biology experts to quantify the nano-architecture of mature and developing sarcomeres.

Activités

The engineer will combine newly developed nanobodies, which recognize domains of Drosophila sarcomeric proteins, with DNA-PAINT super-resolution microscopy to study sarcomeric nanoarchitecture, in particular the structure of Z discs, M bands, and A bands. This analysis will be extended to sarcomeres in developing flight muscles and appropriate sarcomeric protein mutants to understand the mechanisms of sarcomere assembly and maturation in flight muscles. The results will be prepared and submitted for publication.

Compétences

The candidate must be fluent in spoken and written English. He or she must have excellent skills in documenting and presenting scientific results at group meetings and seminars. He or she must be flexible and able to plan his or her schedule independently, and be highly motivated to pursue scientific studies. In-depth knowledge of super-resolution microscopy, particularly the DNA PAINT technique, is a major asset. In-depth knowledge of Drosophila genetics and single-molecule imaging data analysis is important. Knowledge of sarcomere and developmental biology as well as physics is an asset. The candidate must have excellent writing skills to draft the planned manuscripts.

Contexte de travail

The IBDM, located on the Luminy Campus, comprises approximately 220 permanent researchers, lecturers, engineers, and technicians, as well as temporary staff on fixed-term contracts, postdocs, doctoral students, and interns, spread across 22 research teams and 11 technical platforms and services. The IBDM is a joint research unit under the supervision of the CNRS and AMU, which explores the field of developmental biology and associated pathologies. The activity will be carried out within the “Muscle Dynamics” team led by Frank SCHNORRER. As the laboratory is composed of several nationalities, the use of English is essential.

Contraintes et risques

The candidate will work with a computer and high-intensity laser microscopes. He or she will use standard chemicals to fix fly tissues.
