In the framework of the ANR project PARASED, the goal of the proposed postdoctoral position funded by the "région bretagne" will be to decipher the plasticity of the virulence factors of highly invasive marine eukaryotic parasites of bivalves belonging to Perkinsus (Perkinsozoa, Alveolata ).
The post-doctoral position will aim: 1) to evaluate whether the targeted transcriptomic methodology is representative of the global expression of the parasitic genes expressed during the infection and 2) to understand how P. olseni can infect two different hosts species (one native and the other one invasive) belonging to the genus, Ruditapes.
Objective 1: Validation of the methodology. The first step will be to validate targeted and global transcriptomic methodologies on culture trophozoite life stages. All generated sequences will be assembled, annotated and incorporated into a metabolic reconstruction using KEGG comparisons to identify probable gains/losses and significant regulation of parasitic metabolic pathways. A comparison of these data sets will reveal the representativeness of the targeted relative to the global transcriptome. The first data of both methodologies are already available for analysis. The post-doctoral fellow will also work in collaboration with A. Monier U. Exeter to create a bioinformatic pipeline that will be used for the analysis of the objective 2.
Objective 2: Analysis of targeted transcriptomes from P. olseni infected gill tissue samples of two Ruditapes host species from bay of Brest. The post-doctoral fellow will have to carry out RNA extractions and set up the template for Illumina sequencing. The bioinformatics analysis will be proceed using the pipeline created in objective 1.
The highly motivated candidate must have defended its thesis for less than 2 years and must have spent at least 18 months abroad. The candidate must have pratical experience and autonomy in molecular biology as well as 'Omics' analysis (bioinformatic and statistical analysis). Finally a good level of English is also required
The position (38h30/week) will be located at LEMAR (PANORAMA), UMR6539 in Brest. For the bio-informatic analysis, the candidat is strongly encouraged to make short stays in Adam Monier's team (Exeter University) in order to complet the high-standart analysis.
