Missions

This postdoctoral position is part of the PEPR B-BEST initiative (Biotechnologies and Bioresources for a Sustainable and Transformative Economy), and specifically the interdisciplinary project PuLCO (“Multidisciplinary approach to copper metalloenzymes for improving the use of recalcitrant polysaccharides”).
The goal of the project is to develop and apply innovative solid-state NMR methodologies—particularly in paramagnetic environments—to elucidate the molecular mechanisms of LPMOs (Lytic Polysaccharide Monooxygenases), key enzymes in the degradation of lignocellulosic biomass.
The candidate will contribute to the structural analysis of the Cu(I)/Cu(II) active sites and their interactions with polymeric substrates, using cutting-edge MAS solid-state NMR techniques (ultra-fast spinning, proton detection, multidimensional experiments), in combination with quantum chemical calculations.

Activités

Implement advanced solid-state NMR experiments for resonance assignment and structural determination of copper-containing proteins.
Study enzyme-substrate interactions in both crystalline and amorphous samples, under diamagnetic and paramagnetic conditions.
Develop tailored pulse sequences for paramagnetic systems (adiabatic irradiation, pulsed recoupling).
Acquire and process high-resolution multidimensional NMR data.
Collaborate closely with project partners (biochemists, modelers, EPR and electrochemistry experts) for sample preparation and data interpretation.
Contribute to the supervision of PhD students and interns involved in related aspects of the project.

Compétences

Solid-state NMR expertise, particularly on proteins or metal complexes.
Strong understanding of paramagnetic effects (hyperfine shifts, relaxation) and their analysis.
Proficiency in ultra-fast MAS (>60 kHz), proton detection, and 3D/4D correlation experiments.
Competence in complex NMR data processing and interpretation.
Experience in chemical modeling or collaboration with theoreticians (e.g., DFT) is an asset.
Ability to work in an interdisciplinary and collaborative research environment.

Contexte de travail

The work will be conducted at the Centre de Résonance Magnétique Nucléaire à Très Haut Champ (CRMN) in Lyon, a joint research unit of CNRS, ENS de Lyon, and Université Claude Bernard Lyon 1 (UMR 5082), internationally recognized for its methodological and experimental contributions to solid-state NMR.
The CRMN hosts high-field NMR spectrometers up to 1 GHz, including state-of-the-art ultra-fast MAS probes (up to 110 kHz), well-suited for the study of paramagnetic systems.
The postdoctoral fellow will join an interdisciplinary team working on the PuLCO project, which brings together partners in biochemistry, biophysics, and modeling to unravel the mechanism of LPMOs.
The CRMN is also actively involved in national (IR-RMN/Infranalytics) and European (PANACEA) infrastructures, providing a dynamic and collaborative environment with broad academic and industrial interactions.
