Job Description

Thesis Subject

Lanthanides (Ln) cover ubiquitous and crucial roles in modern technology, including for green energy transition, optics, medical diagnostics and radiation therapy. In this context, a recently discovered (2018) Ln binding protein, Lanmodulin (LanM), is of interest for emerging applications in biometallurgy and bioimaging due to its outstanding Ln coordination properties (e.g. high affinity and high selectivity vs Ca2+ and d-block metals). However, LanM applications are still hampered by low kinetic inertness and low intra-Ln selectivity. In order to overcome these limitations and unlock LanM full potential for (bio)technological applications, the OPTIMAL project aims to develop engineered LanM variants with optimised coordination properties. To this end, unprecedented chemical modifications, notably non-native amino acids (nnAAs) for Ln coordination as well as for advanced characterization (e.g. by 19F-NMR), will be introduced in LanM via a semi-synthetic approach based on native chemical ligation (NCL). This method will allow the modification of a protein fragment with nnAAs and its ligation with the unmodified complementary fragment, which will be recombinantly expressed. As a result, OPTIMAL will deliver artificial Ln-binding sites with high affinity and intra-Ln selectivity, boosting the potential of LanM and Ln-proteins for relevant societal applications. In addition, OPTIMAL will also broaden the fundamental knowledge on the coordination properties of Ln-binding proteins, which is currently limited, significantly contributing to this blooming research field.

Your Work Environment

The work will be carried out at the Laboratory of Coordination Chemistry of the CNRS
(UPR 8241) in the "Alzheimer, Amyloids & Bioinorganic Chemistry" team under the supervision of Dr. Enrico Falcone and Dr. Christelle Hureau (team leader).
