Description du sujet de thèse
Regulation of gene transcription is the principal mechanism underlying appearance of the drug-tolerant, dormant, forms of bacteria and the chronic latent infections like tuberculosis. Current project aims to understand molecular mechanisms of transcription termination in the human pathogen, Mycobacterium tuberculosis and its impact on antibiotic resistance.
The study will be focused on the structure-functional analysis of the transcriptional factors (Tfs) binding to RNA polymerase (RNAP) of Mycobacterium tuberculosis. An objective of the project is to identify novel partners binding to Tfs and RNAP by using proteomics approaches. In addition, function of these factors in elongation and termination of transcription will be explored using a combination of biochemical methods, genetics and fluorescent reporters in the live cells.
Contexte de travail
The thesis will be carried out in the team of Konstantin Brodolin at the Infectious Diseases Research Institute of Montpellier (IRIM, Montpellier, France). IRIM is located in the CNRS - University of Montpellier campus which offers a rich and dynamic scientific environment. The team explores molecular mechanisms of transcription regulation in pathogenic bacteria using biochemical, biophysical and structural approaches.
