Description of the thesis topic

This PhD project aims to understand how the protein KIF2C, a microtubule depolymerase, regulates the fidelity of cell division by forming biomolecular condensates at kinetochores during mitosis. These condensates, which are dependent on the kinase PLK1, appear to be essential for correcting erroneous attachments between microtubules and chromosomes, a critical mechanism to prevent chromosomal instability commonly observed in many cancers. The PhD candidate will investigate the mechanisms of KIF2C oligomerization, identify specific partners within the condensates, and analyze their role in mitotic spindle organization, microtubule stability, and activation of the spindle assembly checkpoint (SAC). The work will rely on advanced cell biology approaches (optogenetics, Halo-tagged cell lines, TurboID), biochemistry, and live-cell imaging. The ideal candidate will hold a Master's degree in cell biology, biochemistry, or molecular biology, with a strong interest in the regulation of cell division and solid hands-on experience in experimental biology. The project will take place in a dynamic and collaborative research environment, involving two research teams, one based in Toulouse and the other in Paris.

Work Context

The Institute of Human Genetics (IGH) is a joint research unit affiliated with the CNRS and the University. It hosts around 220 staff members and is organized into three departments. The PhD student will join the team “Spatial Control of Genome Integrity and Oncogenesis”, under the supervision of Dr. J. Basbous (DR2, CNRS). IGH is located on the Arnaud de Villeneuve campus and is part of the Objectif Employeur Pro-Vélo program, which promotes sustainable commuting.
