Job Description

Thesis Subject

Across all stages of health and in pathophysiological conditions, changes in gene and protein regulation drive changes in cell state to allow for diversification of physiology required in multicellular life. These changes are driven by the local cellular environment which, in the case of multicellular tissue, includes mechanical and biochemical cues from surrounding cells as well as the surrounding extracellular environment. How local environmental cues, which operate through mechanotransduction and biochemical signaling networks at seconds to minute time scales, alter transcriptional changes at long time scales is not well understood. The PhD candidate will develop theoretical and data analysis method to interpret single-cell data from endothelial tissues and to produce a working model of how phenotypical changes occur over long time scales. To do this, we will build on a landscape picture of stochastic gene expression dynamics inferred from data using modern machine learning techniques. The results will inform us about how processes on different timescales coordinate to result in the diversity of observed cellular responses.

Your Work Environment

The student will work in the group of T. Mora and A. Walczak that works on various questions of theoretical biophysics, as well as in the broader biophysics groups at LPENS and at the ENS bio institute. As part of our collaboration, the group of Margaret Gardel at the University of Chicago will produce single-cell expression data from endothelial tissues to understand how the cell's transcriptome impacts the morphological dynamics.
