Job Description
Thesis Subject
The thesis subject concerns the development and use of site-selective modification methods in order to generate antibody-drug conjugates (ADCs) with a high control of the drug to antibody ratio (DAR), as well as exploring innovative immuno-conjugates.
Employing both organic synthesis and bioconjugation skills, the heart of the project will concern the synthesis of know or new click functions and disulfide rebridging agents. The latter will be adapted to a solid support context (on-support disulfide rebridging), to afford a single-site selective modification process, of which iteration will give access to a wide range of controled DAR.
In parallel, the disulfide rebridging method will be combined to enzymatic digestion to generate clickable antibody fragments. The latter will be chemically assembled to generate new antibody formats.
The immuno-conjugates generated during the PhD will be submitted to biological evaluation, in which the PhD might be involved if interested.c
The recruited person will carry out steps of organic synthesis and chemobiology (chemical modification of proteins), purification (normal and reverse phase, exclusion chromatography, affinity chromatography...), and analyses (NMR, mass spectrometry, SDS-PAGE gel). The desired profile should have a good theoretical and practical background in organic synthesis. Knowledge in chemobiology/biology would be a plus but is not mandatory. We are looking for someone motivated, curious, sociable, and meticulous.
Your Work Environment
The PhD will be hosted in the SMP group (Systèmes Moléculaires Programmés), member of the OrgaSynth Team, at the IC2MP institute (Poitiers, France).
The group is focused on smart molecules and biomolecules applied to the diagnosis and treatment of diseases, especially cancer, through the development of Vector/targeting systems, triggered release, protein derivatization, cell therapy...
The team is currently composed of 3 permanent researchers (S. Papot (Prof, Head of the team), I. Opalinsky (CNRS researcher) and F. Thoreau (CNRS researcher)), 3 post-docs and 6 PhDs.
