Description of the thesis topic

Chaperones are a class of proteins participating to protein folding that are present in every organisms. One of this chaperone, Hsp90, is highly conserved from bacteria to human. Hsp90 is essential in some bacteria including the aquatic model bacteria Shewanella oneidensis, and it participates to the virulence of many pathogenic bacteria. However, only few proteins that are assisted by Hsp90 (called client proteins) have been identified in bacteria. Hsp90 collaborates with the DnaK chaperone and a functional link has been established with the HslVU protease. The aim of the thesis project is to better understand the mechanism of action of the Hsp90 chaperone in bacteria by identifying Hsp90 client proteins and by defining how Hsp90 integrates in the proteostasis network (made of chaperones and proteases). Several approaches including molecular microbiology, genetics, biochemistry and proteomics will be implemented. This project should give a better understanding of how Hsp90 works in bacteria on a fundamental aspect. In the future, it could lead to new strategies to limit the virulence of some pathogenic bacteria.

Work Context

The PhD student will work at « Laboratoire de Bioénergétique et Ingénierie des protéines (UMR 7281) » in the team « Adaptation systems of bacteria ». The laboratory is part of « Institut de Microbiologie de la Méditerranée (Campus CNRS Joseph Aiguier, Marseille) ». The PhD student will be under the direction of Olivier Genest, CNRS researcher. The candidate will be hired as part of a collaborative ANR project. The team has several collaborations at national and international levels. The PhD student will have to register at « Ecole Doctorale des Sciences de la Vie et de la Santé (ED62), Aix Marseille Université ».

Constraints and risks

The candidate should be highly motivated and have a good knowledge in fundamental microbiology, molecular biology and biochemistry. An aptitude for teamwork is essential.
