Description of the thesis topic

Aggressive tumors are characterized by a metabolic profile often associated with the Warburg effect. Metabolic communication and competition between tumors and immunity cells contribute to inhibiting immune responses by energy exhaustion in tumors (Naquet, 2016). We have shown that the energy level of sarcomas is improved when pantheinase Vnn1 is expressed (Giessner, 2018), limiting the Warburg effect and promoting immune signature. The objective of this project is to show that pharmacological conditioning of tumors by products of pantheinase activity can restore the anti-tumor immune response and increase the efficacy of immunotherapy targeting inhibitory molecules such as CTLA4 or PD1.
This project will involve the manipulation of tumor models in mice, coupled with a study by cytofluorometry, mass cytometry, transcriptomic and metabolic analysis of infiltrating immunocytes and a functional analysis of the anti-tumor potential of immunity effectors in vivo and in vitro. A good knowledge of anti-tumor immunology is required, coupled with a molecular and cellular expertise of these effectors.

The candidate will be in charge of managing independently the research missions for which he/she is responsible. The candidate will work under the responsibility of a Professor and a Study Engineer.

o The use of tumor models in mice
o Comprehensive phenotyping of the mouse immune system by multiparameter flow cytometry.
o He (or she) will perform cellular and molecular immunology experiments
o He/she will participate in collective interest functions in the research team and write technical protocols and reports on the different experimental approaches
