Job Description

Thesis Subject

Nutrients derived from food fulfill three major functions: structural (building), energetic, and regulatory. While the first two are well established, the regulatory function remains largely unexplored. In particular, the nutrients that exert regulatory roles, as well as their mechanisms and sites of action, are still poorly characterized.

Recent work from our team has revealed a direct link between circulating citrate, a metabolite derived from dietary intake, and male gametogenesis in Drosophila. We discovered that the conversion of citrate into acetyl-CoA controls the stability of key proteins involved in spermatogenesis through N-terminal acetylation. Remarkably, this metabolic regulation affects only a subset of proteins and is not observed in the female germline.

The objective of this project is to broaden our understanding of the specific mechanisms that couple nutrition, metabolism, and cellular differentiation. Using Drosophila spermatogenesis as a model system and combining genetic and biochemical approaches, this project aims to: (i) elucidate the specificity of metabolic regulation through the model of N-terminal acetylation, and (ii) identify novel metabolic pathways involved in the control of male fertility.

The planned experiments will include genetic approaches, tissue dissections, Western blot analyses, immunostaining, and confocal microscopy. This integrated strategy will provide a detailed understanding of the molecular mechanisms linking environmental factors—particularly nutrition—to male fertility.
