Job Description
Thesis Subject
-------- MERCI DE TRADUIThis fundamental project at the intersection of biology and chemistry aims to characterise the function of condensates in B lymphocytes. Condensates are membrane-less structures found within the nucleus, formed through a process known as liquid-liquid phase separation (LLPS). They enable certain proteins and nucleic acids (DNA or RNA) to be temporarily and locally aggregated in a specific region of the nucleus to meet a cellular requirement. The role of condensates in the 3D organisation of the genome is thus crucial for chromatin remodelling and the regulation of gene expression. In murine B lymphocytes (B cells), immune system cells that produce antibodies, our experiments have shown that proper three-dimensional (3D) organisation of the nucleus is essential for controlling the immune response and for preventing the occurrence of genomic abnormalities (mutations and/or oncogene translocations) that give rise to lymphomas. The aim of this project is to characterise condensates and their nucleic acid content in order to understand their role in the 3D organisation of the nucleus and in maintaining the genomic integrity of murine B cells. This project will be carried out in collaboration with established partners: Maggy Colas from the IRCER laboratory (Limoges) and Thierry Forné from the IGMM laboratory (Montpellier).
Your Work Environment
The CRIBL laboratory is part of the GEIST institute, which brings together all the research teams active in the fields of Biology, Health, and Chemistry-Environment in Limoges.
Focusing primarily on the study of B lymphocytes (in both their physiological and pathological aspects), our CRIBL laboratory is one of the key players in immunology in France. CRIBL utilises modern molecular and cellular biology technologies to address questions:
– Fundamental: the physiology of normal B cells regulating antibody production, the study of genetic remodelling associated with B-cell differentiation and maturation, and the biology of B-cell lymphomagenesis.
– Applied to issues in experimental medicine: development of mouse models of immune and onco-haematological diseases, participation in clinical research (via the National Reference Centre for Immunoglobulin Deposition Disorders), and the establishment of new diagnostic tools for lymphomas.
– Finalised: with approaches aimed at improving the diagnosis and treatment of tumour or dysimmune disorders.
