Description of the thesis topic
Epithelia are cell layers critical for achieving vital physiological functions. The loss of epithelial integrity can therefore result in pathologies or letality. Some epithelial cell types (external or involved in key exchanges with the external environment) are strongly exposed to pollutants.
Metallic pollutions result from various human activities and are a major problem in all ecosystems. But the impact on the whole fauna and marine ecosystems is far from being well described. Ecotoxicological studies often focus on commercial species because of risks for human health. While it is well known that metals trigger oxidative stress in organisms, the cellular and molecular mechanisms disturbed are often insufficiently understood.
The PhD projects aims at understanding the impact of chosen pollutants on cellular and molecular processes, the project will focus on:
1) Epithelial integrity (monitored by epithelial-mesenchymal transition, epithelial protein localization, wound healing and regeneration efficiency)
2) On the homoscleromorph sponge Oscarella lobularis chosen for ecological, evolutionaty and practical reasons
3) Mainly on mercury, lithium (and cadmium) because the literature mention their impact on the disregulation of two key signaling pathways (Wnt and Notch)
We are searching for a highly motivated and enthusiast student interested in both cell mechanisms and environmental questions. The applicant must have obtained a master degree in biology or ecology and should have a strong background in several domains (chemistry, ecology, cell and molecular biology) and be highly interested and motivated by transdisciplinary approaches.
The project is a transdiciplinary project. Its fruitful management and realization relies not only on the control and the implementation of various concepts and techniques (chemistry, cell biology, ecology and evolution) but also on the ability of communication of different institutes (mainly IMBE and IBDM , and also MIO and CEREGE) with complementary skills. The student will be directed by Carole Borchiellini, Emmanuelle Renard and André Le Bivic and will benefit of an ad hoc Phd committee composed experts in ecotoxicology and chemistry.
The IBDM (http://www.ibdm.univ-mrs.fr/fr/) is located on the Campus Luminy. The IMBE (https://www.imbe.fr/) is dispatched on 6 sites, The PhD student will mainly be located at the marine station (Endoume 13006) but will have to go to other sites (notably Luminy) for experiments (chemistry, imaging) and for meetings.
Both the IBDM and IMBE are under the control of two institutions : CNRS and Aix-marseille university.
Constraints and risks
- Manipulating dangerous and toxic chemicals
- Frequency of trips from one site to another
- The follow up of some experiments may need to work (from time to time) durong the week ends.
