Description du sujet de thèse

Within the ICB lab, the student recruited (M/W) will be in charge of the synthesis and characterization of inorganic iron oxide NPs. He/she will also be in charge of the surface modification of these NPs with heterobifunctional polymers (ensuring colloidal stability and biocompatibility), multifunctional peptides (helping the passage of the BBB and targeting mitochondria) and a fluorophore. He/she will perform a very thorough characterization of these nanohybrids: TEM, DLS, XPS, SIMS, TGA, SEM, XRD, IR and Raman spectroscopies, zeta potential, BET. An important point will be the fine characterization of the surface chemistry with adapted tools available within ICB.
Within LuMIn, the PhD student (M/F) will be in charge of measuring the impact of an inflammatory state on the relative positioning of PER and MITO and on intraneuronal transport, in the human model SH-5YSY and in human neurons derived from iPSC. He/she will then be trained in cell culture, STED and TEM super-resolution imaging techniques, as well as in the analysis of videomicroscopy data for the extraction of endosomal transport parameters. He (she) will also be able to participate in experiments conducted within the LBM and LBHE.

Contexte de travail

Thesis obtained in the framework of the 80PRIME 2023 action. The student will work in collaboration with 3 other laboratories: the LBM of Sorbonne University to participate in the synthesis of innovative peptides, the LuMIn laboratory of ENS Paris Saclay for the study of intracellular trafficking by microscopy techniques and the LBHE lab to study the passage of BBB.
