Description du sujet de thèse
Secondary DNA structures, alternative to the double helix, not only play a role in DNA topology but are also likely to play a regulatory and functional role in mammalian genomes. This is particularly true for G-quadruplexes (G4s), which form stable single-stranded DNA in vitro. Our laboratory has recently shown that these structures are important for gene expression at the promoter level by promoting chromatin opening, in particular through the use of a new technique enabling them to be mapped in cellular context. The aim of this thesis is to highlight the role of G4s in the insulation of chromatin domains. It will test the hypothesis that G4s can help separate topological domains or active and inactive domains of chromatin. To do this, functional genomics approaches as well as genome editing techniques will be used. This work will be carried out in collaboration with a team in Gif-sur-Yvette specialized in the study of insulation.
Contexte de travail
The student will be supervised by Jean-Christophe Andrau at the Institut de Génétique Moléculaire de Montpellier (IGMM), and will have access to a number of technological platforms. He/she will also have the opportunity to train in bioinformatics in the laboratory and in a workshop organized by the laboratory. We are looking for a motivated and enthusiastic candidate with solid experience in at least one of the following areas: molecular biology, functional genomics, bioinformatics, as well as good knowledge of chromatin and eukaryotic transcription.
