Description of the thesis topic
The objective of this thesis project is to use our technological lead in imaging coupled with electrophysiology to revisit LTD. In particular, we will redefine the respective changes in the organization and dynamics of AMPARs in synaptic transmission during the early and sustained phases of LTD. This will pave the way for new approaches to control LTD. In parallel, we will describe the structural plasticity induced by LTD. We will then determine the molecular link between these two plasticities.
- Use our super-resolution imaging techniques in living and fixed tissues to characterize the organization and dynamics of AMPARs at the nanoscale;
- Combine calcium imaging to highlight active synapses with super-resolution imaging and electrophysiology to correlate the content of synaptic AMPARs with the strength of the associated synaptic response;
- Utilize a recently characterized molecular tool that allows the immobilization of AMPARs both in brain slices and in vivo;
- Develop a long-term fluorescence imaging technique to monitor structural plasticity in both cell culture and brain slices.
Created on January 1, 2011, the Interdisciplinary Institute of Neuroscience (IINS) is a joint research unit attached to the CNRS and the University of Bordeaux.
With 160 people in 14 teams, IINS combines multidisciplinary skills to study the molecular and cellular mechanisms involved in brain activity. The Institute develops innovative methods in imaging, chemistry, physiology and computer science. As a component of Bordeaux Neurocampus, IINS is one of the major actors of the neuroscience community in Aquitaine.
