Deciphering the biased signaling of a GPCR by integrative structural biology

Your Work Environment

Applications are invited for a full-time PhD position (36 months) at the Centre de Biologie Structurale (CBS) in Montpellier. The position is funded by ANR and will be carried out under the supervision of the project scientific manager.

This PhD project aims to decipher how the Crip1a protein modulates CB1R signaling through long-range allosteric interactions and in synergy with specific lipids, namely PUFA-PL. This modulation alters the receptor's conformational landscape and thereby determines the selectivity towards specific Gαi protein subtypes.

Building on recent advances in protein purification ad specific labelling with non natural amino acids, the project will explore the details of the interaction at atomic resolution, including the full length receptor. The goal is to understand the structural and dynamic properties of the complex, the impact of the lipid environment and this will be addressed using advanced NMR methods. and integrative modelling, providing real- insights into conformational changes governing CB1R selectivity.

This project offers the opportunity to contribute in the long term to the rational design of selective compounds of pharmacological interest.

Constraints and risks

We are looking for highly motivated recent Master 2 students with expertise in areas such as:
-structural biology, especially NMR
-protein purification.
