Job Description
Thesis Subject
The microtubule cytoskeleton exerts multiple functions in neurons that need to be continuously regulated to ensure neuronal homeostasis. The structure and function of microtubules are regulated by tubulin post-translational modifications. Polyglutamylation is a modification that is highly enriched in neuronal microtubules. Our team has shown that polyglutamylation controls axonal transport and interactions between microtubules and various microtubule-associated protein (MAPs). One of the MAPs sensitive to polyglutamylation is tau protein, implicated in the pathogenesis of neurodegenerative disorders called tauopathies. But the specific causal relationship and molecular mechanism between microtubule polyglutamylation and tau pathology have not been clarified.
In this project, we will address this issue by characterising novel murine models already available in our lab, investigating effects on primary neurons derived from those mouse models, and analysing patient material. We will also explore whether changing polyglutamylation levels by manipulating polyglutamylating enzymes can mitigate the degenerative phenotypes.
Overall, this study aims to decipher the role of tubulin polyglutamylation in the genesis of tau protein-related neurodegeneration. Our results will lay the ground for the development of therapeutic strategies for microtubule-related neurodegenerative diseases and tauopathies
Your Work Environment
This project will be carried out in the Institut Curie, and the student will have free access to all available core facilities and technology platforms in Orsay and Paris. For this project in particular, we will use the transgenic mouse facility, imaging facility, and the L3 laboratory. All equipment and technology needed for this project is in place, and the methodology is firmly established in our team.
Our team has received diverse funding including from the ERC or ANR that will fully cover the expenses of the planned research throughout the entire period of the thesis. We also plan to apply for funding of a project partially overlapping with this proposal to France Alzheimer programme.
