Missions
The 24-month post-doctoral contract is funded by the ANR. The project aims at developing new antibacterial compounds by targeting the hydrophobic pocket of the replication ring. To this end, the project follows two strategies developed simultaneously and in combination, both supported by innovative preliminary data: (1) a fragment-based drug design (FBDD) approach and (2) a targeted covalent inhibition (TCI) approach. The missions attached to this position concern the production and purification of biological molecules, the analysis of their interactions with specific ligands (SPR, ITC, QMS, SwitchSense), the structural analysis of the complexes formed (crystallogenesis and structure resolution), the evaluation of the antibacterial effects of the ligands developed (penetration, quantification, MIC, MCB...). The missions will be carried out in close collaboration with the project partners.
Activities
Apply (or improve) the protocols already developed (cloning, production, protein purification, crystallization, etc.)
Design and develop experimental protocols necessary for the project
Carry out different types of experiments (molecular biology, biochemistry, crystallography, microbiology, etc.).
Analysis and valorization of the results (reports, presentations, posters, articles), particularly for participation in conferences.
Bibliographic and technological watch
All the work is done in accordance with the rules of scientific ethics and health and safety.
Skills
PhD
Experience in molecular biology and biochemistry, knowledge or expertise in structural biology and/or microbiology appreciated.
Computer skills (Pack office, specialized software (ccp4, Phenix, ImageJ, etc...)
Good command of English
Organizational skills, rigor, autonomy, adaptability, dynamism, being a strength of proposition, interpersonal skills, and taste for teamwork.
Work Context
The laboratory "Architecture and Reactivity of RNA" (UPR 9002 of the CNRS) is located on the central campus in Strasbourg in a diversified scientific environment, both in the institute and on the campus of Strasbourg and beyond, on the Upper Rhine basin. The team "Structure and dynamics of biomolecular machines" (https://ibmc.cnrs.fr/laboratoire/arn/equipes/structure-et-dynamique-des-machines-biomoleculaires/) is interested in the structure-function relationships of RNA and the mechanisms of antimicrobial resistance, using and developing a wide range of biophysical, biochemical and microbiological approaches.
