Missions
The research engineer will be part of the "Cellular functions of ubiquitin-dependent proteolysis" team and will be in charge of studying the mechanism of degradation of the Arabidopsis AGO1 protein by the F-box protein FBW2. The candidate will define the “degron” necessary for the recognition of AGO1 by FBW2, will carry out ubiquitylation assays of the AGO1 protein by FBW2, will characterize the topology of the ubiquitin chains formed and will identify the pathway of degradation (proteasome versus autophagy). The study will include protein half-life measurements, in vitro ubiquitylation assays, analysis of mutants in the autophagy pathway and observations of subcellular localization of FBW2/AGO1 by confocal microscopy.

- Engineering of tools to monitor the half-life of the AGO1 protein
- Purification and characterization of ubiquitin chains generated by FBW2
- Analysis of the subcellular localization of FBW2 and AGO1.
Compétences
The post-doctoral fellow must have a good training in plant molecular biology, in plant genetics, in biochemistry or cell biology. Other essential criteria for this position are strong motivation, autonomy, thinking and problem-solving skills, excellent communication skills in English and a strong work ethic. Previous experience in the field of protein degradation will be a plus. Fluency in reading, writing and speaking English, level B2 according to the Common European Framework of Reference for Languages will be required.
Contexte de travail
The research work will be carried out at the Institute of Molecular Biology of Plants (IBMP) in Strasbourg. The candidate will have the opportunity to interact with another team involved in the project located at GMI in Vienna (Austria).

Usual constraints and risks of a plant molecular biology project. A short mobility (1 or 2 weeks) at the GMI (Austria) could be considered.
