Job Description
Missions
The postdoctoral researcher will take part in the ERC Synergy project 3Stops2go, which brings together four teams around the engineering of alternative decoding for therapeutic purposes. The project developed at I2BC pursues two complementary and clearly identifiable objectives. The first is to design, implement and analyse a large-scale screen in human cells in order to identify novel suppressor tRNAs able to read through premature termination codons (PTCs) with high efficiency and, above all, with good selectivity towards native stop codons. The second is to elucidate the mode of action of TLN468 and its derivatives, a readthrough inducer identified by the team, more efficient than gentamicin and endowed with an unusual selectivity regarding the amino acid incorporated at each stop codon, but whose precise molecular mechanism remains to be established. The successful candidate will contribute to key deliverables of the project, including the design of quantitative reporters, the implementation of functional screens, the analysis of high-throughput data (NGS, ribosome profiling) and the mechanistic interpretation of the results.
Activity
The activities carried out in the course of this project will be:
- Molecular biology approaches (cloning, expression, in vitro transcription and translation).
- Biochemical approaches (protein purification, luciferase assays).
- Human cell culture, handling of lentiviruses.
- Development of a lentivirus-based screen for tRNA optimisation.
- Presentation of the work at team meetings and to our collaborators.
Your Profil
Skills
Essential criteria are a PhD in molecular biology, biochemistry or a related field, solid expertise in RNA and translation, together with a command of cloning, human cell culture, flow cytometry and NGS data analysis. Experience with lentiviral vectors, functional screens or ribosome profiling would be a significant asset. The position requires autonomy, rigour, initiative, an ability to work collaboratively and a good command of scientific English.
Your Work Environment
The position is based at I2BC (CEA, CNRS, Université Paris-Saclay, Gif-sur-Yvette), within the Génomique, Structure et Traduction (Genomics, Structure and Translation) team, which specialises in translation, ribosome dynamics and stop codon readthrough. The postdoctoral researcher will have access to the institute's core facilities (sequencing, flow cytometry, mass spectrometry, cryo-EM) and to the Paris-Saclay research environment, as well as to the collaborations of the ERC consortium.
