Job Description
Missions
Plasma cells (PCs) represent the terminal stage of B‑cell differentiation. B cells are continuously produced throughout adult life and undergo various genetic rearrangements associated with immunoglobulin maturation, including VDJ recombination, class‑switch recombination (CSR), and somatic hypermutation (SHM). Secondary lymphoid organs can also harbor different oncogenic events which, depending on their mechanisms, may initiate oncogenic transformation leading to multiple myeloma (MM) or diffuse large B‑cell lymphoma (DLBCL). We have shown that PC generation can be modeled in vitro by recapitulating the steps associated with PC maturation in the different organs/tissues in vivo. In this project, we aim to build on our preliminary results and develop a CRISPR–Cas9 screen to explore the molecular events involved in tumorigenesis, with a focus on genomic instability and epigenetic deregulation. We will seek specific therapeutic vulnerabilities to target tumor cells. The results of this project should provide key insights for targeting the cellular progenitors of these diseases.
