Job Description

Missions

Liver fibrosis is abnormal scarring of the liver in which healthy tissue is replaced by a rigid and dysfunctional extracellular matrix (ECM). It results from chronic damage, activating myofibroblasts, the main agents of fibrosis, accompanied by an inappropriate immune response. It causes approximately 500,000 deaths per year and can progress to other chronic liver diseases, including cancer. Restoring homeostasis in fibrotic livers is a major unmet medical need at present. Project description: cold Atmospheric plasma (CAP), a weakly ionized gas, is an innovative therapeutic strategy. We hypothesize that CAP can restore tissue homeostasis in the fibrotic liver by modulating myofibroblasts and macrophages via the production of reactive oxygen and nitrogen species (RONS). By inducing post-translational modifications (PTMs) in ECM proteins, CAP could modulate chemotaxis and immune and cellular responses. An international consortium has been created, bringing together four teams: Eq1 (plasma physics), Eq2 (liver biology), Eq3 (3D liver models), and Eq4 (Germany, redox chemistry, and proteomics).
The objectives are to: - Develop a plasma jet to treat pathological liver tissue in situ, and analyze the physicochemical properties of plasma; - Study the pathophysiological processes induced by RONS in models of liver fibrosis, using molecular and cellular biology approaches; - Analyze the MPTs of proteins subjected to CAP treatment using advanced mass spectrometry techniques. Impact and benefits This project could improve understanding of CAP-induced tissue remodeling and pave the way for new therapies for hepatic fibrosis.

Activity

- Establish a genetically modified mouse model of liver fibrosis. - Manage animal breeding and genotyping. - Conduct cold plasma treatment experiments in the animal facility and perform post-treatment tissue analysis. - Treat human and mouse cell cultures in vitro. - Receive training in the use of the cold plasma source from physicists. - Liaise with our German colleagues regarding the shipment of biological samples and sample analysis.

Your Profil

Skills

As part of an ANR project, we are looking for a postdoctoral researcher. The PhD should preferably be obtained in 2025, with in-depth knowledge of liver physiology and the pathophysiology of liver diseases. Experience in the following areas is highly desirable: - animal experimentation (regulatory training at the designer level) essential - human and animal cell culture - biochemistry (WB, immunolabeling) and molecular biology (RT-QPCR) The candidate will be required to work as part of a team with our French and German physicist colleagues.

Your Work Environment

The new multidisciplinary unit NABI (Nanomedicine, Extracellular Biology, Integratome, and Innovations in Health), located on the Saint Germain Campus of Paris Cité University (45 rue des Saints Pères, 75006 Paris) was created on January 1, 2025, and brings together around 70 physicists, biologists, pharmacists, engineers, and doctors, including around 30 permanent staff, under the supervision of the CNRS, INSERM, and Paris Cité University. NABI's scientific objectives are to develop new therapies based on biological (extracellular vesicles) or synthetic (activatable metal nanoparticles) nano-objects for the delivery of personalized therapies for difficult-to-treat diseases. Another aspect of NABI is to develop innovative physics-based therapies such as nanohyperthermia or cold atmospheric plasma to treat fibrotic diseases. The agent will join the emerging Innovative Cancer Treatment team.
