In biological systems, redox (electron transfer) membrane proteins contribute to the maintenance of key biological functions (respiration, immunity, photosynthesis...). We are interested in those which, by their function of generating reactive oxygen species, contribute to the immune and inflammatory response. The elucidation of the molecular mechanisms that allow cells to precisely control electron transport is therefore of utmost importance, both for the primary motivation of the research and for the development of innovative therapeutic strategies.
The objective of the research project is to develop genetically and chemically modified membrane redox proteins for time-resolved spectroscopic studies supported by molecular modeling. The work is part of an ANR program offering a unique consortium of interdisciplinary and fundamental research including experimental and theoretical approaches.
- Optimization and development of membrane protein production
- Purification (detergent or not) and reconstitution in membrane systems (liposomes, nanosystems)
- Chemical and genetic modification of proteins
- functional characterization of modified enzymes
This will allow progress towards the first spectroscopic measurements of transmembrane electron transfer.
We are looking for:
- A talented and enthusiastic PhD or engineering degree holder with a strong interest in membrane proteins.
- The candidate must be self-motivated, creative and able to work in a team.
-Experience in molecular biology and membrane protein expression and/or membrane biochemistry would be highly appreciated.
- Previous research experience in yeast (Pichia pastoris) expression system, biochemical functional analyses and chemical modifications of proteins is not mandatory but will be a plus.
The work will be carried out at the Institut de Chimie Physique (ICP, UMR 8000). ICP is a research unit of the CNRS and the University Paris Saclay. It is located in the university campus of Orsay (south Paris area), easily accessible with RER (Orsay- Ville station). Our laboratory offers excellent material conditions to develop this collaborative project since other partners are in the close vicinity. The laboratory is equipped with standard biochemistry and molecular biology rooms and L2 safety level labs dedicated to cell culture experiments. It also offers an easy access to core facilities (imaging, cytometry and analytical chemistry). The work is part of an ANR program offering an interdisciplinary and fundamental research consortium including experimental and theoretical approaches.
The position is funded by the Agence Nationale de la Recherche (ANR) for a period of 18 months and available starting in January 2022. The position will remain open until filled.
