Bioenergetics and Protein Engineering (UMR7281)
Team: 'Structural flexibility of biomolecules in complex environments'

Activity
The goal of this research project is to carry out a structural characterization of a membrane protein belonging to P.aeruginosa Type 2 Secretion Systems through innovative EPR experiments. In a first part, a protocol concerning the expression via DNA-recombinant methods and the consecutive purification with liquid chromatography of the system of interest will be set up. Successively, different EPR experiments with spin labelled variants will be carried out in vitro to check the impact of conformational dynamics on the protein functionalities. These types of experiments will be performed finally also in native conditions in bacterial cells (in-cell EPR) developing an innovative protocol base on the integration of cutting-edge genome-editing techniques with intracellular labelling reactions. All together the results will allow to elucidate at molecular level the conformational changes occurring in the system during secretion mechanism not only in simple buffers but also at physiological conditions.

Your Work Environment
The « Structural flexibility of biomolecules in complex environment » research team, directed by Dr Elisabetta Mileo, is part of the « Bioénergétique et Ingénierie des Protéines » laboratory at CNRS in Marseille. This group mainly apply SDSL reactions and EPR experiments to investigate the role of structural flexibility and conformational transitions on protein functionalities. Additionally, we aim to develop also innovative strategies to perform these types of spectroscopic experiments not only in vitro, but also in real cellular environments (in-cell EPR). Our teams is currently investigating a broad panel of biological systems, ranging from bacterial chaperones to large multi-protein complexes.
The research unit is collaborating with different teams at national & international levels in order to integrate EPR results with other biophysical techniques (i.e. NMR, X-ray crystallography, SAXS, Cryo-EM ; etc.) and/or computational simulations (i.e. molecular dynamics) in order to elucidate the structure-function relationship for several types of proteins.
