Job Description

Missions

The aim of this postdoctoral project is to determine the role of the NR2F1 gene, a key regulator of brain development, in human telencephalon development and neuronal circuit formation, with a particular focus on the balance between excitation and inhibition.
The candidate will investigate the effects of NR2F1 loss of function using 2D and 3D differentiation models of human induced pluripotent stem cells (hiPSCs) to generate dorsal glutamatergic and ventral GABAergic identities. The generation of assembloids will allow the study of neuronal migration and circuit formation. Single-cell RNA sequencing (scRNA-seq) and electrophysiological recordings using multi-electrode arrays (MEAs) will be used to characterize the molecular and functional alterations associated with NR2F1 loss of function.
This work will help elucidate the mechanisms through which NR2F1 loss of function disrupts brain circuit development and function, and contributes to alterations in the excitation/inhibition balance implicated in neurodevelopmental disorders.

Activity

The successful candidate will work under the scientific supervision of M. Bertacchi and the head of the research team and will:

• Adapt and optimize the methodological approaches of the project in consultation with the supervisors;
• Conduct experiments;
• Analyze and communicate data, including maintaining laboratory records and presenting results during project meetings;
• Follow relevant health and safety regulations;
• Maintain an up-to-date knowledge of the scientific literature;
• Supervise students (Master's or PhD);
• Write one or more research articles in English.

The candidate will use a broad range of molecular and cellular approaches, from human iPSC culture and differentiation in 2D and 3D to phenotypic analysis by immunostaining, imaging, and gene expression analysis by qRT-PCR and single-cell RNA sequencing. These approaches will be applied to control cells and cells with NR2F1 loss of function, differentiated into brain organoids and assembloids.

Experience in the following areas would be particularly appreciated: expertise in 2D or 3D differentiation protocols to modulate the regional identity of neural cells along the dorsoventral (D/V) axis, in particular to generate ventral GABAergic populations; and experience with organoid culture on MEA (Multi-Electrode Array) platforms for recording spontaneous electrophysiological activity.

Your Profil

Skills

• PhD in neuroscience, developmental biology, molecular biology, genomics, or a related field;
• Expertise in cellular and molecular biology, as well as fluorescence and confocal microscopy;
• Expertise in developmental biology and cell culture;
• Experience in electrophysiology using MEA platforms;
• Strong organizational and planning skills, creativity, and a strong sense of responsibility;
• Motivation, perseverance, autonomy, rigor, and ability to work as part of a team;
• Good oral and written communication skills in English (knowledge of French is not required).

Your Work Environment

The research team is located in the heart of Nice and is part of the Institut de Biologie Valrose (iBV), which brings together 27 research teams, approximately 300 people, and 25 nationalities. iBV is an international research center bringing together teams with complementary expertise and a shared interest in translating fundamental research into knowledge relevant to clinical applications.
