Missions

The project concerns the development of a database for medical diagnosis/prognosis by high-throughput multiplexed characterization of the effects of ERG ion channel variants. In this collaborative project and affiliated to the IPMC laboratory, the recruited person will be in charge of the evaluation of the impact of mutations of this potassium channel on its protein structure. The candidate will be in charge of the development of a bioinformatics approach for the structural analysis of the ERG channel, including a scoring system of the impact of genetic mutations of this channel in the patient. Based on crystallization data and open source computational tools, he/she will develop a methodological approach allowing its use by non-experts in a clinical context.

Activities

- Taking charge of the structural part of the project
- Improvement of the existing tools
- Automation of the score evaluation concerning the impact of mutations on the structure of the ion channel.
- Scientific surveillance in litterature for the improvement of the statistical significance of the proposed tool
- Regular presentation of the evolution of the project to the collaborators.

Skills

1. Expertise in the software required for structural analysis (pyMOL, UCSF CHimera, GENO3D, Alphafold ...).
2. Programming skills in R, PYTHON.
3. Expertise in the fundamentals of chemistry and biostatistics.
4. Good knowledge of membrane protein biology.

Work Context

Position available at the Institute of Molecular and Cellular Pharmacology (IPMC) (https://www.ipmc.cnrs.fr/cgi-bin/site.cgi). This mixed research unit under the dual supervision of the CNRS and the Université Côte d'Azur is located on the Sophia-Antipolis university campus in the commune of Valbonne (France). As part of a project funded by the ANR (CarDiag) and in collaboration with the Thorax Institute in Nantes (France), the person recruited will work in the team "Physiology and pathophysiology of ion channels", headed by Florian Lesage, under the responsibility of Dr. Franck Chatelain.
