Job Description

Missions

Histology is commonly practiced in hospitals and involves taking a small tissue sample (biopsy), which is sliced, stained, and ultimately examined by a specialist to diagnose its cancerous nature. This process is time-consuming (1–24 hours) and hinders optimal patient management during surgery or endoscopic exploration.
Recently, we developed a wide-field coherent Raman microscopy technique using speckle illuminations and Coherent Anti-Stokes Raman Scattering (CARS) contrast to produce chemical images of biological samples [1]. However, this technique cannot generate histological images of biological tissues because the CARS contrast is affected by non-resonant background noise, which degrades the chemical images of CH₂ and CH₃ bonds required for constructing histological images [2].
This Postdoctoral research project aims to develop a new technology based on photo-thermal stimulated Raman scattering. This effect has been validated in a "point scanning" modality, where excitation beams are focused on the sample. Here, we propose to investigate the possibility of extending this photo-thermal stimulated Raman mechanism to wide-field imaging using speckle illuminations.

Activity

The work program will focus on three objectives:
1. Simulate photo-thermal stimulated Raman contrast and assess its implementation in wide-field imaging with speckle illuminations.
2. Implement a proof-of-principle experiment demonstrating wide-field photo-thermal stimulated Raman imaging on test samples (liquids, polymer beads).
3. Explore the feasibility of wide-field photo-thermal stimulated Raman histology
