Missions
The candidate will develop cutting-edge analytical methods for single-cell proteomic analysis with a spatial dimension. He/She will design a sample preparation protocol suitable for samples obtained through laser microdissection on a microfluidic system, as well as ultra-sensitive and high throughput LC-MS/MS analysis methods.
Activités
- Develop sample preparation protocols for samples obtained via laser microdissection on a CellenOne-type microfluidic instrument.
- Develop ultra-sensitive nanoLC-MS/MS acquisition methods compatible with the analysis of single-cell protein content using Q-TOF (Bruker TimsTOF Ultra) and Orbitrap-Astral (Thermo) nanoLC-MS/MS systems.
- Evaluate innovative nano-chromatography systems to enhance analysis sensitivity while increasing throughput.
- Optimize MS/MS data processing methods and statistical analyses for single-cell data, leveraging innovative AI-based algorithms.
- Present and promote the obtained results.
- Ensure traceability of analyses within the quality management framework of LSMBO (ISO9001 environment).
Compétences
General, Theoretical, or Disciplinary Knowledge :
Expertise in bioanalytical chemistry,mass spectrometry, and proteomic analysis. Proficiency in bioinformatics tools for MS/MS data processing.
Operational Skills :
- Master proteomic analysis strategies, particularly for ultra-sensitive analysis.
- Be proficient in liquid chromatography techniques (reverse-phase chromatography) and tandem mass spectrometry.
- Be skilled in computational tools for LC-MS/MS data processing.
- Knowledge of microfluidics and cell sorting would be an asset.
- Familiarity with programming languages such as Python and/or R would also be advantageous.
- Manage work within an ISO9001-certified environment.
- Communicate and engage scientific exchanges with project collaborators.
- Work effectively in a team and collaborate in a multidisciplinary context.
- Be proficient in written and spoken English (at level B2 according to the Common European Framework of Reference for Language) for presenting results at conferences and writing publications.
Contexte de travail
The Hubert Curien Pluridisciplinary Institute (IPHC), a joint research unit under the supervision of CNRS and the University of Strasbourg (UMR7178), is a multidisciplinary laboratory where research teams from different scientific backgrounds (ecology, physiology and ethology, chemistry, and subatomic physics) develop high-level research programs, with scientific instrumentation as their foundation. The IPHC is structured into four departments and has a total of 393 staff members, including 257 permanent employees (119 researchers and faculty members, and 138 engineers and technicians), 46 fixed-term contract employees, and 102 PhD students.
The selected candidate will join the LSMBO team (Laboratory of Bioorganic Mass Spectrometry) at IPHC, where he/she will have access to state-of-the-art LC-MS/MS instrumentation for proteomic analysis. The project will be conducted in collaboration with the "Membrane Dynamics & Virus" team led by Dr. R. Gaudin (IRIM, Montpellier) and the Toulouse and Grenoble sites of the national proteomics infrastructure ProFI (UAR2048).
