Job Description

Missions

Photosynthetic processes rely on the function of multimeric complexes. Coordination between the expression of genes encoding their subunits and their assembly is essential for cellular homeostasis. In photosynthetic eukaryotes with chloroplasts, particularly those of the green lineage, a mechanism has been identified whereby the translation of many chloroplast genes is regulated based on their assembly status, enabling spatiotemporal control of the expression of subunits encoded in different genetic compartments (nucleus versus chloroplast). This process has been termed the CES process. In cyanobacteria, at the origin of chloroplasts after the primary endosymbiosis, genes are organized into operons. It is not known whether a similar process of translational control based on assembly status operates in these organisms. This project aims to investigate the evolutionary origin of the CES process. Building on the laboratory's experience in characterizing the CES process in chloroplasts and understanding the biogenesis of the Rubisco complex, the objective of this project is to create and characterize Rubisco mutants in the cyanobacterium Synechocystis sp. PCC 6803 in order to test how the expression and assembly of the complex are coordinated.

Activity

Main Activity: Generate and characterize Rubisco mutants in the cyanobacterium Synechocystis sp. PCC 6803 using the following techniques:
1- Molecular biology (cloning, plasmid mutagenesis, nucleic acid preparation and analysis, PCR)
2- Molecular genetics of cyanobacteria (mutagenesis, transformation, study of gene expression)
3- Biochemistry (Western blot, FPLC)
Secondary activity: Participate in the maintenance of a collection of Chlamydomonas reinhardtii strains.

Your Profil

Skills

Knowledge
- Strong knowledge of molecular biology
- Experience in the genetics of microalgae or photosynthetic bacteria is a plus but not essential

Skills
- Proficiency in cloning techniques and genome editing technologies
- Proficiency in PCR techniques
- Ability to culture cells (plant or non-plant) and work under a laminar flow hood in sterile conditions
- Ability to design and implement experimental protocols
- Ability to analyze and present information (maintaining a lab notebook, preparing summary tables of results, preparing oral presentations).
- Prepare articles for publication in peer-reviewed international journals

Soft Skills

The applicant must hold a degree in engineering or a master in Life Sciences.

Your Work Environment

The “Institut de Biologie Physico-Chimique” (IBPC), founded in 1930 by the Edmond de Rothschild Foundation, is dedicated to multidisciplinary research in all fields of biology. The candidate will join the “Photobiology and Physiology of Plastids and Microalgae” laboratory (UMR7141) led by Dr. Angela Falciatore. The laboratory has about twenty permanent staff members (researchers, faculty members, engineers, technicians, and administrative staff) and about fifteen temporary staff members, doctoral students, and postdoctoral researchers. This laboratory, dedicated to the study of light-induced processes (photosynthesis and photoperception) and chloroplast biology, addresses key questions regarding the biology, evolution, and ecology of microalgae, focusing on various model organisms (Chlamydomonas reinhardtii and diatoms) and ecologically important phytoplankton species, studied using ecophysiological, biophysical, biochemical, genomic, and genetic approaches. The contract is for a non-renewable term of 13 months, funded by Sorbonne Université—Emergence EvoCyanoCES project.
