Job Description
Missions
Type IV secretion systems (T4SS) are bacterial nanomachines involved in the transfer of genetic material and intercellular communication. They play a major role in the dissemination of antibiotic resistance. This project aims to obtain high-resolution structures of the T4SS pilus by cryo-electron microscopy (cryo-EM), with a clear objective of rapidly generating structural data suitable for publication.
Activity
The postdoctoral researcher will play a central and autonomous role in driving the project:
i) Implement established protocols for the production and purification of bacterial pili
ii) Ensure quality control of samples using biochemical and structural approaches (western blot, negative-stain EM)
iii) Prepare samples for cryo-EM (grid preparation, optimisation of vitrification conditions)
iv) Interact with cryo-EM facilities for data acquisition
v) Contribute to the analysis of structural data
vi) Contribute to the dissemination of results (rapid publication expected)
Your Profil
Skills
Qualifications:
PhD in microbiology, biochemistry or structural biology
Postdoctoral experience is desirable
Essential skills:
Strong expertise in microbiological techniques
Proven experience in the purification and analysis of protein complexes
Experience in structural biology, ideally in cryo-EM
Desirable skills:
Experience in cryo-EM image analysis
Experience with complex bacterial systems (pili, flagella, secretion systems)
Personal qualities:
High level of autonomy
Strong experimental rigour
Curiosity
Ability to work effectively in an interdisciplinary team environment
Your Work Environment
The project will be carried out at IGDR (Rennes), within the T4-SECRET team, under the supervision of Kévin Macé.
The scientific environment includes:
i) microbiology and biochemistry platforms (BSL-1/2, AKTA systems)
ii) access to a local cryo-EM microscope
iii) high-performance computing resources
iv) access to state-of-the-art facilities (ESRF, HPC Jean-Zay)
The team is young, dynamic, and strongly focused on integrated approaches in structural biology. This project builds on a key experimental barrier that we have overcome, enabling the rapid generation of high-impact structural data on bacterial secretion systems.
