Job Description

Missions

Cell surfaces display a large number of oligosaccharides which, depending on their structure, play a key role in immunity, adhesion, and cell–cell communication processes.
The ability to modify cell surfaces with well-defined natural oligosaccharides therefore offers highly promising opportunities to target or confer new properties to viral and cellular vectors in the field of gene and cell therapy. However, the vast majority of complex natural sugars are not commercially available.
This research project aims to synthesize natural oligosaccharides (heparin-derived structures and oligomannosides) capable of interacting with specific cellular receptors.
Once synthesized, these ligands will be grafted onto viral vectors (AAVs) used in gene therapy in order to improve their tropism and infectivity.
Bioconjugation will be carried out using a click-electrochemistry strategy developed within the group, enabling the rapid decoration of bacteria and living cells within minutes without affecting their viability [Nat. Commun., 2023, 14, 5122; Chem. Sci., 2021, 12, 15374–15381; J. Am. Chem. Soc., 2018, 140, 17120–17126].

The recruited postdoctoral researcher will primarily be responsible for the synthesis of specific natural oligosaccharides and will also perform their functionalization with bioorthogonal groups activatable through electrochemical methods.

Activity

-Multi-step synthesis of natural oligosaccharides
-Electrochemical bioconjugation reactions
-Characterization of synthetic products and conjugates
-Preparation of scientific reports
-Presentation of research at conferences and scientific meetings
